Забор крови в микроветты, содержащие ЭДТА, производили у пяти здоровых доноров при профилактических осмотрах. В кровь in vitro добавляли раствор NaNO2 для получения метгемоглобина (MetHb). В качестве антиоксиданта использовали комплексный препарат, состоящий из следующих активных компонентов: янтарная кислота, инозин, рибофлавин и никотинамид. Измеряли спектр поглощения суспензий эритроцитов с различным содержанием препарата Dι (λι )exper с шагом 1 нм. Методом нелинейной регрессии рассчитывали концентрации производных гемоглобина в суспензиях.</p></sec><sec><title>Результаты</title><p>Результаты. В экспериментах, когда метгемоглобин взаимодействовал с препаратом, оптическая плотность пиков, характерных для оксигемоглобина, увеличивалась, а спектральный пик метгемоглобина снижался. Чем больше были концентрация препарата и время инкубации, тем эффективнее был процесс восстановления метгемоглобина до оксигемоглобина.</p></sec><sec><title>Заключение</title><p>Заключение. Экспериментально доказали, что при начальной концентрации метгемоглобина в крови 91—93% добавление препарата снижает его концентрацию до 25—27%. В то же время, благодаря автовосстановлению, концентрация метгемоглобина уменьшается только до 84%. Показанный эффект может иметь практическое применение при критических состояниях, при хранении донорской крови, при проведении гемотрансфузии, при воздействии физико-химических факторов на кровь. </p></sec></abstract><trans-abstract xml:lang="en"><p>The purpose of the study — to determine the feasibility of using the succinate-based antioxidant for the in vitro reduction of excessive methemoglobin to oxyhemoglobin in blood.</p><sec><title>Materials and Methods</title><p>Materials and Methods. Blood sampling was performed in five healthy donors in microvettes containing EDTA during prophylactic examinations. NaNO2 solution was added to blood samples in vitro in order to yield methemoglobin (MetHb). The complex drug containing the following active ingredients: succinic acid, inosine, riboflavin, nicotinamide, was used as an antioxidant. The absorption spectrum of red cell suspensions with different drug content Dι (λι )exper was measured with 1 nm increments. The non-linear regression method was used to calculate concentrations of hemoglobin derivatives in suspensions.</p></sec><sec><title>Results</title><p>Results. In our experiments, when methemoglobin reacted with drug the optical density of peaks typical for oxyhemoglobin increased and the spectral peak of methemoglobin decreased. The greater the concentration of drug, the more was the incubation time, the more efficient was the process of reduction of MetHb to HbO2.</p></sec><sec><title>Conclusion</title><p>Conclusion. We proved experimentally that while the baseline concentration of MetHb was an average of 91—93%, addition of drug decreased its concentration to 25—7%. Without drug, due to autoreduction, the concentration of MetHb decreases only to 84%.
