Одним из таких подходов является генная терапия, направленная на активацию эпикарда, что обеспечивает образование васкулогенных клеток-предшественниц и секреторную поддержку «сборки» сосудов de novo.</p><p>Цель — изучить возможность активации клеток эпикарда и постинфарктной васкуляризации сердца путем введение генетической конструкции, кодирующей PDGFBB.</p><sec><title>Материал и методы</title><p>Материал и методы. В работе использована модель экспериментального инфаркта миокарда у крысы с последующим интрамиокардиальным введением физиологического раствора, контрольной плазмиды и плазмиды, кодирующей PDGFBB. Исследование влияния PDGF на активность клеток эпикарда выполнено на модели ex vivo, а также клеточной культуре мезотелия in vitro.</p></sec><sec><title>Результаты</title><p>Результаты. Постинфарктное введение плазмиды, кодирующей PDGFBB, способствует увеличению плотности сосудистой сети в периинфарктной области, а также миграции перицитов в зону повреждения. PDGFBB способствует активации пула клеток эпикарда, повышению экспрессии в них маркеров гладкомышечных клеток (показано на модели ex vivo) и появлению признаков активации эпителиально-мезенхимального перехода (in vitro).</p></sec><sec><title>Заключение</title><p>Заключение. Внутримиокардиальное введение генетической конструкции, кодирующей PDGF, после экспериментального инфаркта стимулировало васкуляризацию периинфарктной зоны, что могло быть отчасти обусловлено активацией эпикардиального пула клеток.</p></sec></abstract><trans-abstract xml:lang="en"><p>Increasing incidence of ischemic diseases and limited resources for their treatment stimulate increased interest in studying the mechanisms of vascularization and finding new approaches for its promotion. One of these approaches is gene therapy aimed at activating the epicardium to produce the vascular precursor cells and microenvironment for the «assembly» of de novo vessels.</p><p>The aim is to investigate the possibility of activating epicardial cells and post infarction cardiac vascularization by injecting a genetic construct encoding PDGFBB.</p><sec><title>Material and methods</title><p>Material and methods. A model of experimental myocardial infarction in a rat with subsequent intramyocardial injection of normal saline solution, control plasmid and plasmid encoding PDGFBB was used. The study of PDGFBB effect on epicardial cell activity was performed on the ex vivo model, as well as in vitro mesothelial cell culture.</p></sec><sec><title>Results</title><p>Results. Post infarction injection of plasmid encoding PDGFBB increases the density of the vascular network in the peri-infarct area as well as migration of pericytes to the injured zone. PDGFBB promotes activation of epicardial cell pool and expression of smooth muscle cell markers in them (shown on the ex vivo model), as well as stimulates activation of epithelial-mesenchymal transition (in vitro).</p></sec><sec><title>Conclusion</title><p>Conclusion.
