Установить влияние гипоксии и интерлейкина-6 (ИЛ-6) на экспрессию клаудина-5, окклюдина-1, рецепторов ИЛ-1 и ИЛ-6 в клетках нейроваскулярной единицы (НВЕ).</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. В исследовании in vitro сформировали клеточную модель НВЕ, состоящую из нейронов, астроцитов и эндотелиоцитов. Провели культивирование НВЕ в условиях нормоксии и гипоксии с содержанием кислорода в среде 15, 10, 7 и 5%, а также с добавлением сыворотки больных из групп с максимальной и минимальной концентрацией ИЛ-6. Инкубацию клеток проводили в режиме нормотермии в течение 30 мин с дальнейшей оценкой экспрессии маркеров повреждения клеток НВЕ.</p></sec><sec><title>Результаты</title><p>Результаты. Уменьшение интенсивности флюоресценции для клаудина-5 наступало при 10% кислорода в среде (15,2 vs 34,3 в контрольной группе (р=0,0105)) и менее. На окклюдин-1 гипоксия не оказала влияния. Увеличение флюоресценции для экспрессии рецепторов интерлейкина-1 отметили при 7 и 5% кислорода (соответственно 12,2 и 12,9 vs 9,9 в контрольной группе, р=0,0105), но гипоксия не повлияла на экспрессию рецепторов интерлейкина-6. В группе с нормоксией и во всех группах с гипоксией добавление сыворотки пациентов значимо влияло на экспрессию исследуемых маркеров, гипоксия потенцировала данные эффекты. Добавление сыворотки с максимальным содержанием ИЛ-6 приводило к наибольшему снижению интенсивности флюоресценции маркеров.</p></sec><sec><title>Заключение</title><p>Заключение. Воздействие ИЛ-6 оказало более выраженное действие на экспрессию исследуемых маркеров в клетках НВЕ, чем гипоксия. Гипоксия с концентрацией кислорода в среде до 10% не обладала действием на экспрессию маркеров.</p></sec></abstract><trans-abstract xml:lang="en"><p>The high incidence of postoperative cognitive dysfunction in children undergoing cardiac surgery underscores the urgent need for effective neuroprotective strategies.</p><sec><title>Aim</title><p>Aim. To examine the effects of hypoxia and interleukin-6 (IL-6) on the expression of claudin-5, occludin-1, and interleukin-1 (IL-1) and IL-6 receptors in neurovascular unit (NVU) cells.</p></sec><sec><title>Materials and methods</title><p>Materials and methods. An in vitro NVU model comprising neurons, astrocytes, and endothelial cells was established. The cells were cultured under normoxic and hypoxic conditions with oxygen concentrations of 15%, 10%, 7%, and 5%. The cultures were also treated with patient-derived sera containing high or low levels of IL-6. All incubations were conducted under normothermic conditions for 30 minutes. Injury marker expression was then assessed using fluorescence analysis.</p></sec><sec><title>Results</title><p>Results. Significant reductions in claudin-5 fluorescence intensity were observed at oxygen levels of 10% and below (15.2 vs. 34.3 in controls, P=0.0105). Hypoxia did not affect occludin-1 expression. IL-1 receptor fluorescence intensity increased under 7% and 5% oxygen conditions (12.2 and 12.9 versus 9.9 in the control group, P=0.0105), while IL-6 receptor expression remained unchanged. In both normoxic and hypoxic conditions, adding patient sera significantly altered marker expression; hypoxia enhanced these effects. Sera with the highest IL-6 levels induced the most pronounced reduction in injury marker fluorescence.</p></sec><sec><title>Conclusion</title><p>Conclusion. IL-6 had a more significant impact on injury marker expression in NVU cells than hypoxia did.
